PRODUCTION, ESTABLISHMENT OF THE STANDARD BACOSIDE A3 AND ASSESSING THE QUALITY OF BACOPA MONNIERI EXTRACTS UNDER DIFFERENT EXTRACTION CONDITIONS

Abstract

Bacoside is the name of triterpene glycosides with dammarane structure, known to be the main chemical constituent in Bacopa monnieri, which possesses many valuable biological activities, especially effects on the nervous system to help improve memory, enhance cognition and learning. Bacoside A3 is one of the major bacosides found in B. monnieri, which is considered to be the main component responsible for the activity of bacosides and has been used as a standard to determine the total bacoside content in

  1. monnieri raw material and extracts. However, currently in Vietnam, bacoside A3 is not available and must be imported from abroad with a very high price and long order time. Therefore, in order to be proactive in creating a domestic source of bacoside A3, in this article we present the results of preparation and establishment of the standard bacoside A3 (with high purity ≥ 98%, yield of 0.006% compared to dry sample) from B. monnieri collected in Thanh Hoa province. Simultaneously, the process of quantitative analysis of total bacosides (the total content of five major bacosides: bacoside A3, bacopaside I, bacopaside II, bacopaside X, và bacopasaponin C) according to bacoside A3 was developed. The analytical procedure had high reproducibility, accuracy and sensitivity, was applied to evaluate the quality of B. monnieri extracts under different extraction conditions. The results showed that the extraction method 5 (soaking dried B. monnieri powder in water at room temperature for 8 hours and then removing water and extracting with alcohol (96 degrees) at 60oC produced the extract with the highest total bacoside content (10.6%). This is the first time that bacoside A3 has been prepared and established as a standard from Vietnamese B. monnieri and was applied to evaluate the quality of B. monnieri extracts under different extraction conditions.
